چکیده

The developmental competence of in vitro-matured (IVM) rhesus macaque cumulus oocyte complexes (COCs) is deficient compared with in vivo-matured (IVM) oocytes. To improve oocyte quality and subsequent embryo development following IVM, culture conditions must be optimized. A series of experiments was undertaken to determine the role of epidermal growth factor (EGF) during IVM of rhesus macaque COCs. The addition of Tyrphostin AG-1478 (a selective inhibitor of the EGF receptor EGFR) to the IVM medium yielded fewer oocytes maturing to metaphase II of meiosis II (MII), decreased cumulus expansion, and a lower percentage of embryos that developed to the blastocyst stage compared with untreated IVM controls, indicating that EGFR activation is important for IVM maturation in the rhesus macaque. However, the addition of recombinant human EGF (r-hEGF) to the IVM medium did not enhance outcome. The expression of mRNAs encoding the EGF-like factors amphiregulin, epiregulin, and betacellulin in cumulus cells indicates that these factors produced by cumulus cells may be responsible for maximal EGFR activation during oocyte maturation, precluding any further effect of exogenous r-hEGF. Additionally, these results illustrate the potential futility of exogenous supplementation of IVM medium without prior knowledge of pathway activity.

P-28: Hydrostatic Pressure Induced Cell Deathin Cumulus Cells and Improved In vitro Maturationof Oocytes from Preovulatory Follicles

Background: Cryopreservation of ovaries is an important technique in assisted reproduction technology. Physical forces like hydrostatic pressure have a pivotal role in reproduction systems. Due to changes in intrafollicular pressure during ovulatory process, this study designed to examine the effects of hydrostatic pressure on oocyte maturation and cell death in cumulus cells from cryopreserved...

P-29: Effects of Growth Factors and GranulosaCell Coculture on In vitro Maturation ofOocytes

Background: The maturation medium for in-vitro oocyte maturation is usually supplemented with serum. However, supplementation of serum from pregnant women adversely affects the outcome of in-vitro maturation. The purpose of the study was to assess if growth factors or granulosa cell coculture could overcome the adverse effects of pregnant women’s serum. Materials and Methods: The basal maturati...

Background: Oocyte in vitro maturation success rate is still low in compareision to oocyte in vivo maturation. Thus prediction and improvement of oocyte competency are two critical issues in assisted reproductive technology. New insights into oocyte-cumulus cell regulatory loop may assist our optimization of in vitro maturation technique. Maturation and developmental competency of cumulus oocyt...

Porcine oocyte maturation in vitro: role of cAMP and oocyte-secreted factors – A practical approach

Polyspermy or the penetration of more than one sperm cell remains a problem during porcine in vitro fertilization (IVF). After in vitro culture of porcine zygotes, only a low percentage of blastocysts develop and their quality is inferior to that of in vivo derived blastocysts. It is unknown whether the cytoplasmic maturation of the oocyte is sufficiently sustained in current in vitro maturatio...

Activation of PKA, p38 MAPK and ERK1/2 by gonadotropins in cumulus cells is critical for induction of EGF-like factor and TACE/ADAM17 gene expression during in vitro maturation of porcine COCs

OBJECTIVES During ovulation, it has been shown that LH stimulus induces the expression of numerous genes via PKA, p38 MAPK, PI3K and ERK1/2 in cumulus cells and granulosa cells. Our recent study showed that EGF-like factor and its protease (TACE/ADAM17) are required for the activation of EGF receptor (EGFR), cumulus expansion and oocyte maturation of porcine cumulus-oocyte complexes (COCs). In ...
